چکیده

Yellow fever virus (Asibi strain) has been grown successfully in insect tissue culture cell lines from the moth Antheraea eucalypti and the mosquito Aedes aegypti. Increases in virus 1iter (MICLD5, of two to three logs were evident in 24 to 72 hours at 26 C in a hemolymph-free medium. Virusinfected A. eucalypti cells remained healthy and in a state of reproduction throughout 12 weeks' incubation, but in A. aegyyti cultures, cell lysis reduced the numbers to 10% of the original count within 120 hours.

Studies on the Physiological Conditions Prevailing in Tissue Cultures

An analysis of some of the physiological factors active in Maitland tissue cultures has been presented in the hope that it may be of some value in clarifying the principles underlying tissue cultures in general. It has been found that the empirically determined necessity of using relatively small amounts of tissue in such cultures is dependent upon the fact that excessive tissue leads to a rapi...

Inhibition by rifampin of African swine fever virus replication in tissue culture.

Vaccinia virus and African swine fever virus are deoxyribonucleic acid viruses of cytoplasmic origin. The fact that rifampin inhibits the replication of the former virus led to an investigation of its effect on African swine fever virus. The virus used was cytopathogenic to a PK-15 cell line, hemadsorbing in pig leukocyte cultures and lethal to pigs. Rifampin clearly inhibited the multiplicatio...

A short treatment of cells with the lanthanide ions La3+, Ce3+, Pr3+ or Nd3+ changes the cellular chemistry into a state in which RNA replication of flaviviruses is specifically blocked without interference with host-cell multiplication

Alpha- and flaviviruses contain class II fusion proteins, which form ion-permeable pores in the target membrane during virus entry. The pores generated during entry of the alphavirus Semliki Forest virus have been shown previously to be blocked by lanthanide ions. Here, analyses of the influence of rare earth ions on the entry of the flaviviruses West Nile virus and Uganda S virus revealed an u...

The Titration of Yellow Fever Virus in Stegomyia Mosquitoes

Titrations were made of yellow fever virus in stegomyia mosquitoes, using rhesus monkeys as test animals. It was found that: (a) The average mosquito immediately after engorging on highly infectious blood contained between 1 and 2 million lethal doses of virus. The titer of freshly ingested blood was as high as 1 billion lethal doses of virus per cubic centimeter. (b) During the fortnight succe...
